An unusual case of paracoccidioidomycosis in Brazil BJM
Macêdo,Danielle Patrícia Cerqueira; Neves,Rejane Pereira; Magalhães,Oliane Maria Correia; Lacerda Filho,Armando Marsden.
This paper reports an unusual case of paracoccidioidomycosis with ulcerations in the head and nose in a Brazilian man. For diagnosis, direct microscopic examination of ulcer secretion and skin samples treated with potassium hydroxide was performed. The isolation of Paracoccidioides brasiliensis occurred at 28ºC and 37ºC on Sabouraud agar with chloranphenicol. Diagnosis was established by the presence of isolated and multiple budding yeast-like cells and typical fungus growth of P. brasiliensis.

Antifungal and antitumor models of bioactive protective peptides Anais da ABC (AABC)
Rodrigues,Elaine G.; Dobroff,Andrey S.; Taborda,Carlos P.; Travassos,Luiz R..
Peptides are remarkably reactive molecules produced by a great variety of species and able to display a number of functions in uni-and multicellular organisms as mediators, agonists and regulating substances. Some of them exert cytotoxic effects on cells other than those that produced them, and may have a role in controlling subpopulations and protecting certain species or cell types. Presently, we focus on antifungal and antitumor peptides and discuss a few models in which specific sequences and structures exerted direct inhibitory effects or stimulated a protective immune response. The killer peptide, deduced from an antiidiotypic antibody, with several antimicrobial activities and other Ig-derived peptides with cytotoxic activities including antitumor...

Takahachi,Gisele; Maluf,Márcia Luzia Ferrarezi; Svidzinski,Terezinha Inez Estivalet; Akimoto-Günther,Luciene Setsuko; Hübler,Márcia Rosangela Neves de Oliveira; Bersani-Amado,Ciomar Aparecida; Cuman,Roberto Kenji Nakamura.
The objective of this work was to evaluate biochemical parameters in Paracoccidioides brasiliensis infected mice and the effect of Canova medicine on these parameters. Mice infected with the isolate Paracoccidioides brasiliensis Pb18 and treated with Canova for 17 weeks were used. The biochemical parameters analysed were the levels of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and amylase, and the concentration of total proteins, albumin and globulins. The results suggested that the animals that were treated with Canova had less functional alterations in their internal organs.

Cell-free antigens from precocious Paracoccidioides brasiliensis culture induce a typical delayed-type hypersensitivity reaction J. Venom. Anim. Toxins incl. Trop. Dis.
CJ,Fecchio; LA,Dias-Melicio; AMVC,Soares; S,Guimarães; MTS,Peraçoli; A,Sartori.
Cell-free antigens (CFAg) derived from Paracoccidioides brasiliensis have typically been used in immunodiffusion reactions for serodiagnosis or therapeutic follow-up of paracoccidioidomycosis patients. Thus, we investigated the usefulness of CFAg obtained from cultures at different ages, to evaluate cellular immunity by the footpad test, in experimental murine paracoccidioidomycosis. Male mice infected with P. brasiliensis 265 strain were challenged in the footpad with CFAg obtained from four- (4d CFAg) or 11-day-old cultures (11d CFAg). The increase in footpad swelling provoked by 4d CFAg and 11d CFAg was similar and showed significant difference in relation to control groups. However, the infiltrate pattern was strikingly different: 4d CFAg induced a...

Central Nervous System Paracoccidioidomycosis: an overview BJID
Almeida,Sérgio Monteiro de.
Paracoccidioidomycosis (PCM) is an infectious disease, endemic to subtropical areas of Central and South America, caused by the dimorphic fungus Paracoccidioides brasiliensis. It is a chronic disease, mostly affecting adult males, with a mean patient age of 44 years. Central nervous system involvement (CNS PCM) has been found in 13% of the patients with systemic disease. We reviewed the clinical presentation, diagnosis techniques and treatments for CNS PCM.

Experimental infections with Paracoccidioides brasiliensis obtained from armadillos: comparison to clinical isolates BJID
Nishikaku,Angela Satie; Peraçoli,Maria Terezinha S.; Bagagli,Eduardo; Sugizaki,Maria Fátima; Sartori,Alexandrina.
Paracoccidioides brasiliensis causes paracoccidioidomycosis (PCM) that is one of the most prevalent systemic human mycoses in Latin America. Armadillos show a high incidence of PCM infection and could, therefore, be a natural reservoir for this fungus. In this study were compared the virulence profiles of isolates obtained from nine-banded armadillos (Dasypus novemcinctus) (PbT1 and PbT4) and isolates from PCM patients (Pb265 and Bt83). Pathogenicity was evaluated by fungal load and analysis of colony morphology. Immunity against the fungus was tested by delayed type hypersensitivity test (DTH) and antibody quantification by ELISA. The higher virulence of PbT1 and PbT4 was suggested by higher fungal load in spleen and lungs. Armadillo isolates and Bt83...

Immunohistochemical demonstration of TGF-ß and decorin in paracoccidioidal granulomas BJMBR
Nishikaku,A.S.; Burger,E..
Different patterns of granulomas have been observed in 6- to 8-week-old mice after ip inoculation with 5 x 10(6) yeast cells of Paracoccidioides brasiliensis. Transforming growth factor-ß (TGF-ß) is a cytokine that has been shown to participate in fibrosis and granuloma formation; its activities seem to be modulated by the small proteoglycan decorin. In the present study, TGF-ß and decorin expression in epiploon granulomas was assessed by immunohistochemistry in susceptible (B10.A) and resistant (A/J) mice after 15, 30, 120 and 150 days of P. brasiliensis ip infection. The epiploon was collected, fixed in Methacarn solution and embedded in paraffin, and 5-µm thick sections were used for immunohistochemical analysis employing the...

Juvenile paracoccidioidomycosis in urban area: report of two cases BJID
Carneiro,Rodrigo da Costa; Miranda,Bianca Grassi de; Camilo Neto,Carlos; Tsukumo,Marina K; Fonseca,Cibele LC; Mendonça,João Silva de.
We present two cases of juvenile form of paracoccidioidomycosis (PCM), a systemic mycosis frequently found in rural areas, whose prognosis is poor in children and young adults. They are a 14-year-old boy and a 25-year-old woman, both residents in an urban area in São Paulo - Brazil, without any history of travelling to an endemic area. They have been admitted to the hospital due to fever, weight loss and lymphadenopathy. The diagnosis was confirmed by serologic and histopathologic study. Patients have recovered after therapy with oral itraconazole and were discharged from hospital, maintaining outpatient visits. In this article, the authors discuss the unusual presentation of PCM in an urban area.

Natural antibodies in paracoccidioidomycosis BJM
Unterkircher,Carmelinda S.; Leão,Mariella V.P.; Blotta,Maria Heloisa S.L.; Camargo,Zoilo Pires de.
Recent attention has been focused on the natural antibodies as a component of natural immunity and as integral part of the idiotypic network. However, their functional role in different infections has rarely been studied. This work was undertaken to investigate the presence of natural antibodies in paracoccidioidomycosis (PCM). In addition, we analyzed anti-P. brasiliensis antibodies and their distribution in IgG subclasses in order to acquire better knowledge about the humoral immune response in this mycosis. Our findings show that the natural antibody response is not very much increased in PCM when compared with other parasite infections and this response is restricted to a few specificities, suggesting that P. brasiliensis moderately triggers CD5+ B...

Standardization and validation of Dot-ELISA assay for Paracoccidioides brasiliensis antibody detection J. Venom. Anim. Toxins incl. Trop. Dis.
Kamikawa,Camila Mika; Mendes,Rinaldo Poncio; Vicentini,Adriana Pardini.
Abstract Background Paracoccidioidomycosis (PCM) is a neglected systemic mycosis caused by a dimorphic fungus of the Paracoccidioides genus. The standard diagnosis is based on isolation of the fungi in culture, and by microscopic visualization of characteristic multiple budding yeast cells in biological samples. However, in some situations, access to the site of injury prevents the collection of biological material. A variety of immuno-serological techniques has proven useful for allowing inferring diagnosis with a certain degree of certainty, thus optimizing time. The aim of this study was to standardize and validate the Dot-ELISA (DE) assay, comparing it with the serological standard, double immunodiffusion (DI). Methods In order to standardize the...

The left lung is preferentially targeted during experimental paracoccidioidomycosis in C57BL/6 mice BJMBR
Tristão,F.S.M.; Rocha,F.A.; Dias,F.C.; Rossi,M.A.; Silva,J.S..
Paracoccidioidomycosis (PCM) is a chronic systemic mycosis caused by the inhalation of the thermally dimorphic fungus Paracoccidioides brasiliensis as well as the recently described P. lutzii. Because the primary infection occurs in the lungs, we investigated the differential involvement of the right and left lungs in experimental P. brasiliensis infection. Lungs were collected from C57BL/6 mice at 70 days after intravenous infection with 1×106 yeast cells of a virulent strain of P. brasiliensis (Pb18). The left lung, which in mice is smaller and has fewer lobes than the right lung, yielded increased fungal recovery associated with a predominant interleukin-4 response and diminished synthesis of interferon-γ and nitric oxide compared with the right lung....

The low efficiency of dendritic cells and macrophages from mice susceptible to Paracoccidioides brasiliensis in inducing a Th1 response BJMBR
Almeida,S.R.; Lopes,J.D..
In the present study we evaluated T cell proliferation and Th lymphokine patterns in response to gp43 from Paracoccidioides brasiliensis presented by isolated dendritic cells from susceptible and resistant mice. T cell proliferation assays showed that dendritic cells from susceptible mice were less efficient than those from resistant mice. The pattern of T cell lymphokines stimulated by dendritic cells was always Th1, although the levels of IL-2 and IFN-gamma were lower in T cell cultures from susceptible mice. To determie whether different antigen-presenting cells such as macrophages and dendritic cells stimulated different concentrations of Th1 lymphokines, the production of IFN-gamma and IL-2 was measured. It was observed that dendritic cells were more...

Thymic atrophy and fungal virulence during experimental paracoccidioidomycosis BJM
Brito,Vânia Nieto; Souto,Paula Cristina de Souza; Gameiro,Jacy; Cruz-Höfling,Maria Alice; Verinaud,Liana.
The immunosuppression observed in systemic mycosis can be related to primary lymphoid organs damage. Thus, our laboratory has studied the effects of the Paracoccidioides brasiliensis infection on the thymus of mice. Here, thymuses of susceptible and resistant mice were evaluated after inoculation with highly and slightly virulent isolates of the fungus. All groups presented thymic atrophy, loss of corticomedullary delimitation and increase of apoptotic index. However, mice inoculated with high virulent strain showed earlier and stronger alterations suggesting that thymic atrophy can be directly related to the fungal virulence and to the immunosuppression.

Validation and clinical application of a nested PCR for paracoccidioidomycosis diagnosis in clinical samples from Colombian patients BJID
Gaviria,Marcela; Rivera,Vanessa; Muñoz-Cadavid,Cesar; Cano,Luz Elena; Naranjo,Tonny Williams.
Paracoccidioidomycosis is a systemic and endemic mycosis, restricted to tropical and subtropical areas of Latin America. The infection is caused by the thermal dimorphic fungus Paracoccidioides brasiliensisand Paracoccidioides lutzii. The diagnosis of paracoccidioidomycosis is usually performed by microscopic examination, culture and immunodiagnostic tests to respiratory specimens, body fluids and/or biopsies; however these methods require laboratory personnel with experience and several days to produce a result. In the present study, we have validated and evaluated a nested PCR assay targeting the gene encoding the Paracoccidioides gp43membrane protein in 191 clinical samples: 115 samples from patients with proven infections other than...
